Liver Fibrosis Model
Balb/c or C57Bl/6 mice
General Model Information
Following academic and industry-accepted best practices, Woodland performs the model as follows: 0.5 microliters CCl4 per gram of mouse body weight is administered IP twice weekly for up to 6-8 weeks in corn or olive oil. Established fibrosis can be investigated after 2–4 weeks of CCl4 treatment, while severe fibrosis (approaching cirrhosis) can be observed after 6–8 weeks of treatment.
Therapeutic dosing
In a typical study, dosing would start after two weeks when fibrosis is fairly well established and continue with both CCl4 administration and the test article for an additional 4 weeks. Usually, sentinel animals are taken down at the time of dose initiation as a baseline reference of disease.
This is preferable over liver biopsy as it is less invasive to the study animals. Survival surgery / biopsy places stress on the animal including the anesthesia, surgical injury, liver regeneration/proliferation with inflammation and requires post-operative analgesia for 3 days, all of which may interfere with therapeutic dosing.

Our CCl4 model conforms to the standard protocols most frequently in use. Custom designed studies are available upon consultation to fit your experimental needs.
Note animal to animal consistency, with a few excursions (deviations) which is why we suggest n=10 animals per group for most studies.
